Area-1255 advice, allopregnanolone for PSSD

I believe strongly in our current theory that PSSD, and PSSD cognitive/memory impairment, is caused by a desensitation of the 5ht1a post-synaptic receptor. This causes influx of the amino acid tryptophan which then interacts with sodium channels that cause the genital anesthesia symptoms. I believe only Lecozotan can cure this, but, despite the pharmaceutical giants spending millions of dollars to put the drug through phase III clinical trials, and the drug showing massive efficacy, it vaporized off the face of the earth under very suspicious circumstances. (possibly because it would undermine all their SSRI cashcow)

Anyway, let me get to my question. I would like to try one of the synthetic 5a-DHP substances by Idealabs as a way to increase Allopregnanolone. I wanted to see if this could possibly benefit the cognitive and memory impairments plaguing me. MY only concern is that the studies are half and half currently.

half the pubmed studies show that it reverses cognitive impairment to an extreme degree and is a miracle wonder.

1. Do you think that this substance can yield benefit for these PSSD issues?
2. why do the pubmed studies conflict? which version is reliable?
3. do you feel the product may be worth trying? or does it increase the risk of advancing the cognitive/memory impairment? is there a risk that it could advance the brain into alzheimers disease prematurely?
4. What is your opinion on this molecule?

Importantly, progesterone decreased responses to faces in the amygdala and fusiform gyrus during memory encoding, whereas it increased hippocampal responses.

Notice, it said Progesterone, it also referred to ''faces'' and emotional recognition, and the 'other half' of your studies mostly involved all females. I don't think that would apply to us, do you?

The rest of the studies are all logical, allopregnenolone enhances GABA-activity, so for some people, that may translate into cognitive impairment, likewise, for us it may have either benefit or cause worsening of Cognition - depending on the person affected.

I think its worth a try. Along with other glutamatergic agents. Most probably Sodium benzoate.

...And its the PRE-SYNAPTIC receptor that is desensitized, not the postsynaptic one, the post-synaptic receptor is causing the body to not be able to synthesize, utilize, or properly activate nitric oxide synthase, in addition to blunting Glutamate, and central dopamine activity.

I guess my Question is, if cognitive impairment did result, would it be long term? or would it advance our acceleration of degeneration to Alzheimer's disease, for us that are already predisposed?

I think you already know the answer to that question.

It's not going to 'accelerate' anything, if it was that bad, it would be pulled off the market.

The majority of Pregnenolone, DHEA, 5-a-DHP users all report spectacularly good results. Probably close to 98% of the users of the former two are impressed through and through. I don't see any like issues from the latter neither.

Please take a look at this study, as it shows that after a month since the administration of Allopregnanolone, there was some significant advencement in the progression of AD.

How come the pubmed test studies conflict each other, one set of studies shows that there a rapid reversal and regeneration with administering it for alzheimers, the other say it advances it; which set of studies is true?

Are you even reading these studies? Clearly, its talking about MICE that ALREADY HAVE AMYLOID PLAQUES. There's no way to translate that study into anything meaningful for humans. Ridiculous.

Also, other neurosteroids compete with Pregnenolone in some cases, and there are naturally occurring hormones that would protect against any even minor, insult that you think would come from Pregnenolone. So, the problem would be gone before it started, unless you are assuming that every human who ingests Pregnenolone has ZERO Testosterone, Estrogen, GABA and etc which would protect against it.

We are wasting time talking about this, its trivial, and has nothing to do with PSSD.

The endogenous neurosteroid allopregnanolone alters neuronal excitability via modulation of the GABAA receptor and causes decreased neurotransmission. In Alzheimer's disease (AD), neurotransmission seems to alter the levels of toxic intracellular amyloid-β (Aβ) oligomers, which are implicated in AD pathogenesis and cause cognitive decline. Inhibition of synaptic activity has been shown to increase levels of intracellular Aβ. Allopregnanolone at endogenous stress levels inhibits synaptic activity and could have similar effects. By using a transgenic AβPP(Swe)PSEN1(ΔE9) mouse model for AD, we observed that chronic allopregnanolone treatment for three months with stress levels of allopregnanolone impaired learning in the Morris water maze. The learning impairment was seen one month after the end of treatment. Chronic allopregnanolone treatment also led to increased levels of soluble Aβ in the brain, which could be a sign of advanced pathogenesis. Since the learning and memory of wild-type mice was not affected by the treatment, we propose that chronic allopregnanolone treatment accelerates the pathogenesis of AD. However, further studies are required in order to determine the underlying mechanism.

I think as far as this Pregnenolone thing, you are overthinking it - you ask good questions sometimes, some of it is due, and some is a little excessive. Still, gotta focus on our objectives here. Gotta be prudent but organized.
